Postdoctoral Research Scientist – Translational Research Institute for Metabolism and Diabetes – Orlando
AdventHealth Orlando seeks to hire a Postdoctoral Research Scientist who will embrace our mission to extend the healing ministry of Christ.
Located on a lush tropical campus, our flagship hospital, 1,107-bed AdventHealth Orlando, serves as the major tertiary facility for much of the Southeast, the Caribbean and South America. AdventHealth Orlando houses one of the largest Emergency Departments and largest cardiac catheterization labs in the country. We are already one of the busiest hospitals in the nation, providing service excellence to more than 32,000 inpatients and 125,000 outpatients each year.
The collaborative, multi-disciplinary team of doctors, scientists, and researchers at the Translational Research Institute for Metabolism and Diabetes (TRI-MD) bridges the gap between discovery research and clinical care. This facility, which houses some of the most technologically advanced clinical and diagnostic equipment available, is dedicated to the study of obesity, metabolism, diabetes, and the metabolic origins of cardiovascular disease. A postdoctoral position is available in the laboratory of Associate Investigator Dr. Paul M. Coen within the Exercise, Aging and Bioenergetics research program. He/she will be a highly motivated individual and will focus primarily on an NIH-sponsored research project grant (R01AG060153) on skeletal muscle atrophy that occurs during bed rest that leads to a loss of muscle strength and physical function. The presence of type 2 diabetes (T2D) may exacerbate this health issue, which represents an enormous public health problem, particularly for older adults, a large proportion of whom have T2D. The overall objective of this project is to decipher the temporal sequence by which mitochondrial dysfunction and lipotoxicity in older adults with T2D impact atrophy and recovery of muscle mass, strength and physical function following bed rest. Studies in the clinically relevant context of aging, T2D and exercise will provide a physiologicalframework in which the temporal and mechanistic relationships between mitochondria, intramyocellular lipid and protein synthesis/breakdown can be interrogated during atrophy and recovery. To address this objective, we will conduct muscle biopsy studies during bed rest to delineate temporal links among early alterations in mitochondria, intramyocellular lipids, and protein synthesis and breakdown networks. We will obtain muscle biopsies before and after bed rest, to determine:
In situ mitochondrial function (respiration, ROS emission, calcium retention) in permeabilized myofibers
Myocellular lipid profiles by 3D shotgun lipidomics
Mitochondrial /Intramyofibrillar fractional protein synthesis/breakdown rates using innovative in vivo D2O-labeling and proteomics
31P-MRS assessments of mitochondrial energetics. The candidate will be capable of working independently and will be responsible for directing research projects, conducting experiments, collecting and analyzing data and writing abstracts and manuscripts for publication.
Monday through Friday; 8a – 5p
